## Description

iNANO at Aarhus University intends to purchase for research purposes a state-of-the art TIRF fluoresccncc microscopc platform for multicolor high sensitivity, low background, single molecule fluorescence imaging. The instrument will be used by multidisciplinary users for multi-channel molecular high resolution fluorescence imaging and spectroscopy, in parcicular, to elucidate correlated molecular behavior involving a number of nucleic acid and protein partners. The goal is to have a platform for multicolor investigations with the possibility of at least 5 lasers for excitation throughout the visible and near infrared range, an active compensation for z-drift for stable long-rime observation, micro and nano positioning stages for precise sample positioning, and flexible multi-channel detection, where the spectral ranges and number of channels can easily be changed bctween 1, 2, 3 and 4 channels. Several TIRF-based approaches and instruments have been weighed and the conclusion is that micro-mirror TIRF uniquely offers a spacial separation of the excitation and emission that will enable multi-color experiments not possible otherwise. To our knowledge, the RM.21 Advanced Microscope is the only instrument on the market that fulfils these requirements.
