- The Imaging facility of Biomedicine wish to purchase a new slide scanner that automatically can acquire immunofluorescence and brightfield images. Specifications: Hardware. The system must be equipped with - Minimum 7 single LED's, be able to handle 100 slides in a single run (the wavelength of the be distributed to cover common dyes in the spectrum from UV ( e.g. DAPI) to far red ( e.g. Alexa Flour 790) - Fast filterwheel for and narrow band filters optimized for the single LED's - 5,10, 20 and 40 x objectives of high quality for fluorescent imaging (minimum Plan-Apochromate), bright field contrast imaging option with a 5 megapixel scientific color camera at 60 frames pr second ( or better) - 12 megapixel monochrome camera with an actively cooled CMOS sensor with a large sensor field of view (minimum 14.1 x 10.4), High dynamic mode and spectral sensitivity in the area from 350 to 1000 nm. Workstation with capacity (storage, graphic card, RAM) to handle a high content imaging system as a slides canner Software: The system must have software that is compatible with our Zeiss LSM800 and LSM900 airy scan confocal microscopes allowing correlative imaging, meaning that is should be able to overlay and organize images form all these sources for multimodal data management. It should furthermore be able to image many slices of different tissue at relatively low magnification and then move them to our high-resolution airy scan confocal, where the software should align and re-find structures of interest for high resolution imaging (approx. 150 nm resolution as minimum in XY dimensions). The acquired images should subsequent be automatically shown in a context where the microscopist can zoom in or out across the borders of resolution domains. Conclusion: This software compatibility is to our knowledge only possible with the Zen Connect model for an AxioScan 7 Zeiss slide scanner, wherefore we wish to purchase such a fully automated slidescanner from Zeiss with the abovementioned specifications.
