The LSM900 Airyscan 2 - Multiplex Mode for Fast and Gentle Confocal Superresolution in Large Volumes The LSM900 allows up to 4x parallelization with Airyscan 2 detector. Imaging of up to 18.9 FPS is possible at increased resolution. The novel detection sampling concept allows rapid volumetric imaging with unprecedented resolution beyond what is available in traditional confocal systems today. Simultaneously, the detector must give an increase in SNR (4-8x). Detector spec. Fill Factor 90% (Fill factor = ratio of a pixel's light sensitive area to its total area). Optimization for scanning speed in super resolution mode must be obtainable.

- The LSM900 is the only CLSM on the market that fulfills our requirements. The microscope will be used by several research groups at iNANO. The high SNR and 90 nm resolution of the Airyscan is a critical for the planned research, which involves visualization of particles and structures on the nm spatial scale in biological samples and nanomaterial samples. Furthermore, the possibility for a subsequent system upgrade with FCS is necessary for future projects in which we study flow in microbial biofilms. ZEISS LSM900 meets these criteria, which are covered by the patents below: EP2317362. This patent describes the unique way of analysing/calculating a super resolution image based on the emission signal acquired with Airyscan. Benefit: Efficient and fast acquisition and display of super resolved images EP2825908. These patents describe the unique detection hardware arrangement of Airyscan. Benefit: Down to 90nm resolution EP3447559. The patent describes the basic principle of Airyscans single elements sub resolution detection method focusing on the usability in 3D. Benefit: Super resolution imaging with Airyscan in 3D EP2122404. This patent describes the collimating optics for each laser being fibre coupled to the scan head to ensure optimal colour correction of the excitation lines along the optical axis. Benefit: Even within the visible range where various fibres are used to couple the visible lasers into the scan head, each fibre or wavelength has a collimating optics. The collimating optics is adjustable but preset. With this correction optics it is facilitated that all wavelengths passing the same AOTF are to hit the same focal plane and there is no colour shift along the optical axis which is important for multicolour imaging. EP2090918. This patent describes the calibration objective used to calibrate the system for constant. performance. Benefit: The objective is a device which is independent of the Z position of the focus drive and when used for the (recommended) calibration procedures, it ensures a constant performance of the system.
